Timing question about a split dose protocol

I want to move my rat up to a higher dosage so that the side effects are gentler on the day of the pin. For those here who have looked into split dosing: what protocol do you use?

Right now the idea is Friday 6pm and Tuesday 6am, which splits it into exactly the same # of hours across the week and keeps the solution concentration steady by spacing the doses at equal times. The problem is that my rat gets sleepy after the Friday pins, so I'm unsure whether adding a Tuesday pin would leave it drowsy on Tuesdays during the day.

Any insights or previous research to share here?

With that approach, would neither one ever reach any sort of steady state? Take TIRZ split dosing as an example — it smooths things out and evens the peaks and valleys. But when you're simply rotating to a different med each time, what impact does that have?

In my case, 5mg is enough for the tirz to do its job....and 5mg is the amount where the reta is effective

I've been on a split dose routine for almost 5 months at this point. My days are always Monday and Thursday, once I'm done with work. It's worked great for me — side effects stay low and my appetite stays under control.

That's interesting. I've also been thinking about simply stacking Tirz with Reta — whatever your split happens to be, say Tirz every 5 days or so — and just dosing some Reta right at the same time, rather than staggering the two.

Each one gets to steady state, just like a standard once-a-week schedule would for either of them.

My own shots go in on Monday evening and Friday morning, though for me the exact timing doesn't matter much.

Splitting has been my approach since December, and I'm down 90lb while splitting.

Out of curiosity, I went looking for any solid data showing how much GLP1, GIP, and Glucagon there is per mg of reta, and how much GLP1 and GIP per mg of tirz.

In short, the idea is to split the GLP1 and GIP across doses while taking glucagon 1 time a week, but I'm really curious about the amounts involved.

Another thing I wonder about: if it were available, could you add glucagon only to tirz and get comparable synergies?

This is just idle curiosity.. but I'd really like to see data

this is what google returns:

Retatrutide does not contain physical percentages or mixtures of GLP-1, GIP, and glucagon.

Instead, retatrutide is a single, unified synthetic molecule (peptide) engineered to mimic and bind to all three of these hormone receptors simultaneously. Therefore, every single milligram (mg) of retatrutide consists entirely of 100% retatrutide. [1, 2, 3, 4]

If your question is regarding its relative potency or binding affinity toward each receptor compared to the body's natural hormones, scientists measure this via EC50 and binding ratios: [1]

    • GIP Receptor: Retatrutide is highly potent here, stimulating this receptor roughly 8.9 times more effectively than our body's natural GIP hormone. [1]
    • GLP-1 Receptor: It acts as a milder agonist, with about 0.4 times the potency of natural GLP-1. [1]
    • Glucagon Receptor: It is structurally tuned to provide a submaximal "gentle" signal, with about 0.3 times the potency of natural glucagon. [1, 2]
This specific structural balance allows the drug to aggressively target fat metabolism and insulin via the GIP/GLP-1 pathways while safely accelerating energy expenditure via the glucagon pathway without over-elevating blood sugar

I take 4mg on Sunday and Thursday, though I prefer to take both doses in the morning since that suits my routine better. It's been working great!

For me, reta and tirz are handled as two separate things....Previously I was taking 5mg of tirz x 2 per week, after which I replaced one of those weekly doses with reta....Tirz gets taken Monday morning, reta on Thursday night. To avoid the supposedly awful sides, my first reta dose was 2mg.....since that reta amount was low, I figured raising tirz to 6mg would even things out. Don't forget, for 4-5 weeks before that I had already been on 5mg twice a week....That 6mg turned out to be past the tipping point, and I ended up with noticeably worse acid reflux, headache, and bad food vibe than I had expected....so on the following one I dropped it back down to 5mg....

Reta, though, gave me no trouble. From the initial 2mg I moved to 3mg once, then spent a couple of weeks at 4mg. 4 was okay, but I was hungrier than I wanted, definitely more so than with tirz...so eventually I moved up to 5mg. At 5 I noticed a little more stomach rumbling, burping...mild headache...plus some 'throat feel' that came close to acid reflux without quite reaching it...and the food noise suppression was roughly equal to the 5mg tirz....with reta it felt pretty much exactly where I wanted to land...

So there's no set schedule, and I don't follow the 'testing group' titration schedule. or wait 4 weeks before changing anything...What I've picked up on is that when my appetite drops, I start burping more, and food stops being at the top of my mind...I'm sitting right on that edge between feeling sort of hungry....and feeling like I might throw up...That's my sweet spot....so to speak 😳...and as things stand with triz and reta, it just happens to be 5mg ea... 😎

I'm sure it will keep shifting, but one nice part of controlling supply, dose, which day, and which compound is that you can wing it and just go with the flow and it will be just fine...👍

This week I'm returning to split dosing. Monday morning I take 1.5 mg of Reta, then Thursday morning 1 mg of Reta. The reason I quit splitting in the first place was that it felt like I was pinning all the time. I've run a few rounds of the Wolverine stack, though, and with that you're pinning daily. So I've realized daily pinning really isn't that rough.

What's your total time on Reta so far? Early on, roughly a day or 2 post-injection, I'd get sleepy, though that faded once a month had passed.

For my Reta protocol, I take 3.5mg on Monday night and another 3.5mg on Friday morning. Without shifting the days around, that's the closest I can get to an even split.

On top of that, when the body is in a caloric deficit it believes it is starving, and the pushback shows up as fatigue, lethargy and feeling cold. Extra protein can help fight those off.

I plan to get my rat to higher dosage to get gentler side effects on the day of pin. For anyone here who has researched split dose: what's your protocol?

At the moment the plan is Friday 6pm and Tuesday 6am to split it at exactly the same # of hours in a week and stabilize the solution concentration by spreading it out at equal times. The issue is my rat feels sleepy after Friday pins so I'm not sure if adding a pin on Tuesday will make it drowsy on Tuesdays during the day.

The three of us — my husband, our son, and myself — pin on wednesdays and saturdays, and the clock time isn't something we worry about. Occasionally one of us forgets and does it a day later, after which we simply return to the regular schedule. Whether it makes any difference, I honestly can't say, although during the first month or two I would feel tired the day after pin day. That no longer happens. Twice a week has been our routine from the start, and we intend to keep it that way unless we reach goal weight, at which point we'd taper down to a maintenance dose purely for convenience.

Which peptide are you using, and how much are you taking per injection?
