I've caught wind of arguments on both sides, though only in broad strokes. On one hand, the trials produced outstanding outcomes when 1 injection was given each week; on the other hand, since the half-life sits at roughly 6 days, effectiveness may taper off toward the end of that window, and dividing the dose brings down peak serum concentrations, which softens side effects.

What I'd really like is for someone to pull these threads together. For example, is a weekly 1-shot regimen the better route for a particular person X or objective A, while splitting doses wins out for a different person Y or objective B? Or am I badly mistaken about all this — if so, I'd be glad to be set straight.

Splitting a dose can make sense for 2 different reasons. One of them is side effects that show up a day or 2 after injecting and then fade for the remainder of the week. That pattern points to peak blood levels right after the dose, so dividing it into smaller, more frequent peaks keeps those side effects away. This tends to help most when you still have weight to lose or you are still hungry on your current dose and want room to go higher, but side effects would otherwise stop you.

The second reason is hunger that only becomes a problem 5 or 6 days out from a dose. In that situation the trough blood level is too low to keep hunger in check, and splitting raises that trough twice a week. If this is your issue, simply raising the dose is the other option.

For the majority of people, the 6 day half life is not something you really feel, since the gap in blood levels between day 5 and 6 is quite small. What once a week really has going for it is simplicity, because it is nearly impossible to stuff up. The more often you dose, the harder it becomes to be certain, is it 2 days or 3 since the last dose? Unless every dose gets written down, a mix-up is likely at some point. For me, every second day dosing or weekly dosing was memorable, but anything in between I ended up messing it up.

Any advice on moving from a split schedule back to 1 injection per week? Like, bump up the initial shot while lowering the later one, step by step, until the whole amount lands on 1 day? Or does that not really matter?

Mostly because I was lazy, I never bothered. Ozempic gave me terrible nausea even at low doses, so I had to dose it 2nd daily to keep that under control. When I later switched to tirz and then added some reta too, I mostly just kept doing 2nd daily dosing out of habit. Your idea of slowly raising one dose while lowering the other seems totally reasonable; making that change all at once could definitely cause problems. Another plus for splitting is when you're titrating upward: if unpleasant side effects hit, the smaller doses usually clear your system quicker than a single larger weekly dose would.

I've been on both sides of this coin. With 1 pin weekly, my results have been phenomenal, even though my desire for food clearly goes up on day 5-6-7. With splitting, results have been mixed. On day 1, I don't care about food whatsoever and have to force myself to eat during days 2-3-4. After that, food starts to become somewhat appealing again, like a tiny, barely noticeable reward — similar to discovering a $5 bill in your pocket that you'd forgotten about while your clothes went through the wash — and I gradually feel more like eating a real meal, until maybe day 6-7 when I actually want 2 real meals, portion controlled but still real. Honestly, it seems to even itself out in the end.

Hey. I'll share what I've been through.

When I first began using tirze for weight loss, I was splitting my doses. At the start, I pinned once a week, on Mondays, at 2.5mg. Once I felt the food noise creeping back toward the end of the week, I chose to add a Friday dose of 1.25mg to cover the weekend.

After that I switched to reta and set up 2 equal doses, on Mondays and Thursdays.

What works for me is the knowledge that peaks and troughs are smaller, which means blood concentration stays more steady.

These days I'm in maintenance on reta, taking 1mg 2 times per week. My weight hasn't moved, my sugar levels are optimal, and there are no sides. Life is good.

Just to clear something up: pinning two times a week really isn’t a hassle to remember. My plan is Monday night and Friday morning, which works out to 3.5 days apart — simple enough.

Neither approach comes with any special trick. Pick whichever you like, and if you later feel like switching, nothing is holding you back.

I hit a plateau while dividing my reta doses at 12mg. Once I switched to a single weekly injection, the weight started coming off again.

For me, injecting once every 6 days works best. Ultimately, it comes down to the signals your own body sends—no one else can make that call for you.

From everything I've gathered, my plan is to switch over to one injection. Appreciate everyone's input and viewpoints.
That's the answer. Should results fall short, switch to pinning 2x per week.

See how the response varies from 1 rat to another in this community?

Pretty crazy, right?

Do you already divide the dose for any other peptides or injections you take? If that’s the case, aligning with that routine is straightforward.

That’s exactly how I handled it—my timing was already set.

My thinking comes down to one thing: smoothing out the highs and lows.

What I can’t work out is why that would get the stall to break.

If you look at it technically, one 12mg shot would hit a higher peak than taking 6mg and then another 6mg 3.5days later. Maybe that higher peak is what set off the signalling.

Personally I split my doses, mainly because I'm working to keep my allodynia under control, and keeping track of which days and times to dose isn't hard for me at all. The steady suppression is something I value too, and I can't picture switching to a single dose unless something about my situation changes.

In my opinion, having appetite return on days 5, 6, and 7 is actually a good thing. That said, it varies a lot from person to person, and the number on the scale when you begin is likely the most important variable.

Could you tell me which dose strengths you went with, how much weight you lost, and over how long?

And besides R, was there anything else you took?

Con 2,5 mg he visto que la supresión del apetito me dura 4 días, mientras que los días restantes paso bastante hambre. Me ronda la idea de fraccionar la dosis en 2 inyecciones de 1,5 mg cada una.

In a span of 6 months, I dropped roughly 24% of my body weight. The first phase was tirze at a low dose of 3.75mg/week, and after 3 months I switched to reta. My reta starting dose was 2x2mg/week. For maintenance at present, I take just 2x1mg/week. Over the past 2 months, I have additionally been on Klow and Nad+, both taken every day.
