Hello everyone — I'm something of a super-newb. California is where I was born and raised, but the UK has been home for coming up on 10 years, and those 10 years have taken a toll on my body, my mind and my overall well-being.

Weight loss is my initial reason for being here. Losing weight used to come easily whenever I decided to. Countless times I dropped 50+ lbs without much struggle. That no longer holds.

Every effort to shed fat, recover my mobility, my confidence and my effervescent personality leaves me drained & exhausted. Which is what brought me to this forum.

Prescriptions for glp1s have sat unused since 5 years ago through December 2025. A straight GLP1 strikes me as unlikely to help, given how little I eat already. The real obstacle is a fatty liver that refuses to budge — it blocks fat burning no matter how strict my eating stays or how often I work out. So the plan here is to research any and all peptides that can drive fat loss first, and ideally the rest of my health too.

One thing I'd especially like to know: whether any particular peptides address compound MTHFR gene mutations.

Glad to be joining this community!

Hey!! Another American based in the UK here. Which area are you in? I'm around Leeds. Regarding MTHFR, I'm homozygous for the c667t variant. From what I've read about peptides, I haven't seen much mention of it—perhaps glutathione is something to look into? Since my mthfr effects seem minimal as far as I can tell, I haven't explored peptides, but I do take methylated b vitamins, steer clear of folic acid, have used TMG, and omega 3 is supposed to be beneficial. HTH.

GLPs could still be beneficial in your case. It's likely that you're dealing with some degree of metabolic dysfunction and insulin resistance, and GLPs are able to assist with those. Reta, in addition, may help by activating the glucagon receptor, which supports burning fat and calories. It's also helpful for fatty liver, though the other options might offer that too, so I'd suggest researching that.

Hey! Meeting fellow Americans over here in the UK is always great. I'm mainly based in Manchester, though I'm in Glasgow quite a bit since that's where my husband comes from. Which area are you in?

Because of my MTHFR mutations, my blood is sticky, and I've had blood clots before. I'll definitely check into glutathione, thank you!

Appreciate it! My worry is that I'd shed what little appetite I do have - junk food goes down with no issue 🤣 Yet my normal diet is fairly clean & getting enough in is a battle.

Someone mentioned combining Tirz with Reta - maybe I ought to look into that arrangement as well. I'm quite willing to give it a try.

Oh wow, blood clots — that's frightening. During my pregnancy I was put on heparin for that reason.

For stronger appetite suppression, the usual combo people run is Tirz alongside Reta. Honestly, Reta sounds like it could suit you better, since it doesn't cut appetite as hard. I'm using it myself and it's worked well for me. Looking back, if I were starting over, I'd likely have gone with Tirz for the appetite suppression — though that's assuming I could stay at a lower dose. As it happens, I'm on a lower dose of Reta and it's been fine (so maybe I'm being silly thinking Tirz would have been better for me…) ! You'll pick up a lot here, and that might make your choice easier. Ultimately, how you'll respond isn't something you can always predict, but at least you can start out informed.

are you saying you were prescribed a GLP 1 while in the UK? Was that through the NHS or private, I didn’t realize they were available for people without type 2 back then!

Nope — that came from my doctor back in the States, though my GP here was willing to take his script and issue me one of her own. GLP1s were never the reason I wanted it; levothyroxine was what I needed. I'm not sure many GPs in this country are as forward-thinking as mine.
