I set out to build a few nasal sprayers for Oxytocin, and the tiny amounts involved caught me off guard. Below is the clinical intranasal dosing I managed to track down:

Daily intranasal amounts fall between 10-48 IU, varying by purpose:

10-20 IU taken once a day for general wellness,

20-24 IU for social anxiety or bonding,

24-48 IU divided into 2 daily doses for autism research protocols, or 40 IU ahead of therapy sessions for PTSD.

Effects begin within 15-30 minutes.

Keep it refrigerated between 2-8°C.

With no preservative present, use it within 48-72 hours, or freeze aliquots to store it longer.

Because intranasal oxytocin contains no preservative, it breaks down fast in the fridge (48-72 hours). Splitting it into individual aliquots and freezing them stretches shelf life considerably — you take out 1 small portion as needed, finish it within 48-72 hours, and leave the remainder frozen. That way a large batch doesn't go to waste, and prepared solution keeps for weeks or months rather than days.

When preparing intranasal oxytocin, reach for sterile saline rather than BAC water.
 
Below is the protocol I've settled on. If you spot any errors, tell me and I'll revise it.

*NOTE: if the bac water stings too much, switch to sterile saline in place of BAC water. The spray simply won't keep as long (72 hours maximum, for safety's sake)

Oxytocin Nasal Spray: Compounding & Dosing Master Guide##

Phase 1: Reconstitution (Initial Mix)

1. Sanitize your work area and swab the rubber stoppers on every vial with alcohol.

2. Pull exactly 3.00 mL of bacteriostatic water into the syringe.

3. Push the liquid in slowly against your 5 mg oxytocin powder vial. Point the flow toward the inside glass surface of the vial.

4. Gently swirl until no powder remains. Never shake.

* This yields a stock solution at 1.67 mg/mL (~833.3 IU/mL).

## Phase 2: Compounding Your First Nasal Spray

1. Take exactly 0.36 mL (36 units on a U-100 insulin syringe) from the freshly mixed stock solution.

2. Put that 0.36 mL into your empty 5 mL nasal spray bottle.

3. Measure exactly 4.64 mL of sterile saline and add it to the bottle, so the total reaches 5 mL.

4. Close the bottle and swirl it gently.

* The nasal spray is now usable, giving ~6 IU per 0.1 mL spray.

## Phase 3: Aliquoting & Freezing the Remainder

1. Take up the rest of the stock liquid into your syringe in separate portions.

2. Move exactly 0.36 mL (36 units) into a clean, empty, sterile 3 mL glass vial.

3. Do this again until you have filled 7 backup vials, each with 0.36 mL.

4. Close the vials tightly and put them directly into the freezer.

## Phase 4: Administration Guidelines

  • Daily Dose: Use a total of 4 sprays per day (24 IU total).
  • Application: Once daily, give 2 sprays into the left nostril and 2 sprays into the right nostril.

## Phase 5: Refreshing Your Bottle

Once your current 5 mL nasal spray bottle is empty:

1. Take one 0.36 mL backup vial out of the freezer.

2. Leave it at room temperature until fully thawed.

3. Pour all of the thawed vial's 0.36 mL contents straight into your empty spray bottle.

4. Add exactly 4.64 mL of saline so the bottle is filled back to the 5 mL mark.
