The calculation itself isn’t hard. Still, if you’d rather skip doing the unit conversions yourself, or you just want to verify something fast, here’s an endotoxin calculator that showed up in Telegram last weekend.

You enter the total endotoxin content per vial, the peptide mass, your dose, and your body weight. It then checks that against the FDA limit.

As an example, I’ve attached a screenshot of the calculation for the recent FSD0706 R30 batch. The test results are real; the dose and bodyweight figures are just random.

Much appreciated for putting together this calculator, and pardon the rookie question: where might someone locate the cutoff thresholds on their own? That way I could run the numbers myself if I chose to?
 
The FDA standard limits are what most people treat as a “cut-off.” Those figures appear both on that calculator page and throughout many forum threads. Here is the wording from the calculator page:

Based on the FDA/USP pyrogenic dose threshold of 5.0 EU/kg/hour for parenteral (IV/SC/IM) administration.

So you can either work it out yourself by calculating backward, or enter your weight and dose into the calculator, then keep raising the endo amount per vial until you reach the FDA limit (or whichever limit you choose based on your own health conditions and personal risk tolerance).

Quick question about how the formula works.

Dose maths at 5.0 EU/kg/hour

= 5.0 EU * 80kg each hour, i.e. 400 EU/hour for 176lb


Shouldn't the uptake be slower than the full 8mg per 1 hour the calculator shows?

A subcutaneous dose of tirzepatide begins absorbing into the body immediately after injection and takes between 8 to 72 hours to reach its maximum peak concentration (\(T_{max}\)) in the bloodstream. According to the FDA official prescribing guidelines from Eli Lilly, the median time it takes to hit this peak is 24 hours (1 day). [1, 2]

Key Absorption Timeline

    • Immediate (0 to 8 hours): The medication enters the fatty tissue layer, starts diffusing into local blood vessels, and begins activating receptors.
    • Peak Window (8 to 72 hours): Blood levels steadily climb, reaching their highest concentration within this timeframe.
    • Average Peak (24 hours): Most individuals experience maximum circulating levels exactly one day post-injection
