A question on filtering hold back.

Hi Everyone,

How do you personally calculate what volumes to use to flush the filter?

I had a thought that I could recon the vial with 2 or 2.5ml and filter that, then use another 1 or 0.5ml to flush the filter and take it up to the desired concentration. The hold back volumes I've seen are much smaller than that though. Is there a disadvantage to doing it this way?

Pretty sure my Biologix 13mm hold .1ml/10units. Easy to test one. This shows how to check volume.
